The aircraft will need to have everything removed down to it's basic empty weight to get the new weight. Balance is calculated from the figures from each load cell.

I was a AMC load planner once upon a time and we weighed each axle and calculated center of mass for cargo. I would assume that weighing an airplane would be similar.
